Bluephage Easy Kit for Enumeration of Somatic Coliphages
Based on the ISO 10705-2 – Single Agar Layer (SAL) method
Bacteriophage: Somatic Coliphages
Method: ISO 10705-2 & ISO 10705-2 & US-EPA 1602, 1642 and 1643
Description
This detection and enumeration kit is based on ISO 10705-2 and US-EPA 1602, 1642 and 1643 methods. It contains all the consumables and biological material required to perform the analysis, including freeze-dried specific host-cells for the somatic coliphage group, which are ready for use after 120 min of incubation.
MSA and MSB ready to use
Host strain (WG5) and positive control included (ɸX174)
Additional material required and not provided in the Kit:
• Sterile Petri dishes (140mm)
Applications of the Easy Kit BP 1604:
The Easy Kit 1604 is designed to specifically detect and enumerate somatic coliphages by incubating a 100 mL water sample with the appropriate host strain. This method applies to samples where low bacteriophage counts are expected, e.g. drinking water and reclaimed water.

What is contained in the Easy Kit BP 1604
It contains all the consumables and biological material necessary to perform the test, including freeze-dried host cells specific for the somatic coliphage group, ready for use after 120 – 150 min incubation.
How to perform the coliphage test with the Easy Kit BP 1604
Mix the 100 ml water sample with a fresh culture of the host strain. Add this mixture to a melted solid nutrient medium and spread in 140 mm diameter Petri dishes.
Incubate the plates, which will be visible after (18±2)h. The results are summed and expressed as the number of plate-forming units (pfu) per 100 ml. > Assay time to results is one day, including incubation for (18±2)h. 8.
Number of tests
This kit is designed to perform 10 assays with one 100 mL water sample in each one.
Application
Samples with expected low counts of bacteriophages (e.g. drinking water, reclaimed water…)
Sample volume: 100 mL
Number of assays: 10 assays
Product code: BP1604
What are coliphage viruses?
Somatic coliphages are bacteriophages of enteric origin that infect Escherichia coli through cell surface receptors. Somatic coliphages in a water sample usually indicate contamination by human or animal feces or sewage containing these excrements.
Why test for coliphages?
Bacteriophages offer complementary advantages to bacterial indicators. They tend to persist longer in the environment, and their resistance in water and food more closely resembles that of human enteric viruses than fecal viruses, thus providing more information about viral pathogens that are not adequately represented by studying bacterial indicators alone. Therefore, they are a relatively quick and simple method for detecting fecal contamination.
What regulations require the analysis of coliphages?
Somatic coliphages are included in water, wastewater, biosolids, and food guidelines and regulations, complementing bacterial indicators such as E. coli and enterococci.
